- Description
- Neuropeptide Y/NPY in Human Brain. The specificity of the NPY antiserum was determined by radioimmunoassay. Synthetic peptides, somatostatin-14, leu-enkephalin, met-enkephalin, oxytocin, Arg8-vasopressin, vasoactive intestinal peptide, angiotensin II demonstrated less than 0.005% cross- reactivity and substance P less than 0.02%. Peptide YY and bovine pancreatic polypeptide demonstrated approx. 36% and 0.05% cross- reactivity respectively. Human peptide YY share 65% homology amino acid sequence for human NPY. Synthetic NPY completely abolished radio labelled NPY binding to NPY antibody in RIA and antibody binding to tissue antigen. Neuropeptide Y/NPY was detected in immersion fixed paraffin-embedded sections of human brain (hypothalamus) using Mouse Anti-Human Neuropeptide Y/NPY Monoclonal Antibody (Catalog # MAB8517) at 15 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS002) and counterstained with hematoxylin (blue). Specific staining was localized to neuronal processes.
