Endogenous agonist at orexin receptors (Ki values are 20 and 38 nM for OX1 and OX2 receptors respectively). Stimulates feeding following central administration and may be involved in the control of sleep-wake cycle and other hypothalamic functions.
Description
Orexin A and Orexin B are neuropeptides produced in the lateral and posterior hypothalmus. They bind the OX1 and OX2 GPCRs which are involved in food uptake. Orexin A binds to OX1 & OX2 with equal affinity while Orexin B primarily binds to OX2. In addition the Orexins have been shown to have roles in wakefulness, mood, addiction, and lipid metabolism. Orexin A, also known as hypocretin 1 (hcrt1), is a 33 amino acid excitatory neuropeptide, localized in a small set of neurons in the perifornical area of the hypothalamus. It is synthesized from the precursor protein prepro-orexin in neurons. Orexin A binds to its G-protein-coupled receptor orexin-1 (OX1) which is also called as hcrt receptors 1 (hcrtr1).
